SDF-1 induces directional chemotaxis of endogenous BMSCs at the intervertebral fusion site and promotes osteogenic differentiation by regulating Wnt/ß-catenin in the bone marrow chimera spinal intervertebral fusion mouse model

Clinical observations show that the current spinal fusion with internal fixation has a nonfusion rate of 5%-35%; however, methods to promote are limited. This study aimed investigate role SDF-1-induced directional chemotaxis BMSCs in bone marrow chimera intervertebral mouse model. were isolated from and identified by detecting CD44/CD34 positive cells. (GFP-BMSCs) labeled GFP for tracking vivo. Mice inoculated GFP-BMSCs construct model, which divided into BM-SIF BM-SIF+SDF-1, BM-SIF+SDF-1-Anta group. The callus area site was detected radiology. HE staining used detect trabeculae formation. Expressions osteogenic molecules fibroblast markers RT-PCR Western blotting. showed obvious adipogenic differentiation ability, according oil-red O alizarin-red staining. Bone model successfully established, efficient localization at site. SDF-1 significantly promoted formation increased transcription/expression graft mice; SDF-1-Anta (AMD3100) decreased transcrition/expression, compared those mice group (p < 0.05). markedly induced proliferations mice, enhanced expression Wnt10b ß-catenin In conclusion, regulating Wnt/ß-catenin pathway modulating proliferation BMSCs.
